      摘要:[目的]为了探明EuREF1与胶分子量和含胶率的关系,深入探究杜仲胶合成机制。[方法]在杜仲新枝和叶片生长较快的4月中旬到6月中旬,利用RT-PCR技术分析杜仲EuREF1基因在雌雄株叶片和茎皮中的表达水平,以索氏提取法和GPC/SEC分别检测叶片和树皮中橡胶的含量和分子量。[结果]在4月至6月期间,雌雄株叶片、茎皮EuREF1表达水平分别与这些器官中胶含量及胶分子量成显著或极显著正相关关系(R=0.898 2~0.988 0)。在雌株叶片、茎皮、果皮和雄株叶片、茎皮中,分子量在1.0×106Da至5.0×106Da 所占比例为最多,即长度在1.5×104至7.4×104个异戊二烯单体的橡胶为最多。[结论]结果推测EuREF1基因产物与杜仲胶积累程度有紧密的关系,可能在橡胶链的延伸中发挥重要作用。

      Abstract:[Objective] To explore the relationship between EuREF1 and molecular weight, rubber content, and to explore the synthesis mechanism of gutta-percha.[Method] From the middle of April to the middle of June, the new stems and leaves of gutta-percha grew rapidly, use RT-PCR technique to analyze the expression level of EuREF1 gene in male and female leaves and stems, and use Soxhlet extraction and GPC/SEC respectively to detect the rubber content and molecular weight of rubber in leaves and stems.[Result] From April to June, the expression levels of EuREF1 in female leaves, stems and male leaves,stems are significantly or highly significantly positively correlated with rubber content and molecular weight in these plant organs (R=0.8982 ~ 0.9880). Among the leaves, stems , fruit of female and leaves , stems of male , The proportion of molecular weight of 1 million KD to 5 million KD is the most, and the rubber with length of 15,000 to 74,000 isoprene residues is the most. [Conclusion] The results suggest that the product of EuREF1 gene is closely related to the accumulation of gutta-percha and may also plays an important role in the elongation of rubber chain.
